Production and optimization of amylase and glucoamylase using Aspergillus oryzae under solid state fermentation

Filamentous fungi have been widely used to produce hydrolytic enzymes for industrial applications. Fungal enzymes amylase and glucoamylase were produced using a specific culture Aspergilus oryzae under solid state fermentation and the enzymes obtained were partially purified using isopropanol. Screening of four agricultural residues viz. rice bran, wheat bran, rice bran: wheat bran (1:1), rice bran: paddy husk (1:1) was done to produce amylase and glucoamylase. From these, rice bran was selected as substrate for further study as it yielded maximum amylase (2.72 IU) and glucoamylase (4.11 IU) activity. Various cultural conditions such as temperature, moisture content, pH, spore suspension and incubation period were optimized. The temperature was varied from 20 to 40 °C, moisture content from 10 to 30 ml / 5 gm, pH from 3 to 7, Spore suspension from 1x10 5 to 1x10 8 spores/ml and incubation period from 3 to 6 days. Temperature of 30°C, moisture content (20 ml/ 5 gm), pH 5.0, spore suspension (1 ml) of 1x10 7 spores/ml and incubation period of 5 days were found optimum for maximum production of enzymes. © 2013 Universal Research Publications.
